Chapter 5, Problem 3Q
Summary Introduction

To review:

Whether something different is learned by researchers by using the method of cell fractionation, which is used to determine, the path of tagged protein.

Introduction:

Cell fractionation is the method, by which cells are fractured or opened so that protein expression of the cellular components can be determined. The cells, which are infected by a fluorescently labeled virus, are first subjected to homogenization so that the cellular components like mitochondria, nucleus, and endoplasmic reticulum come inside the homogenization mixture.

The homogenized mixture is then checked for the expression of proteins at different times. The green fluorescent protein (GFP) is also used to check for the presence of different proteins in the cellular components.

It is desired to isolate genomic DNA from liquid culture of S. cerevisiae yeast. A commercial kit will be used to isolate genomic DNA from this liquid culture. Answer the following questions to understand the strategy used by commercial kits for genomic DNA isolation. a)      List all the steps from cell pellet preparation to DNA elution. b)      With which feature can the membrane in the column that comes with the commercial kit bind DNA? c)      Which component in the kit would you use to recover the DNA from the membrane of the column to which the DNA was attached?
Diagram and explain how APEX probes can be used to determine that an individual is CC (homozygous) for a specific G/C SNV. Recall that the genotype of an SNV is identified from the strand shown in NCBI. What color fluorescence will be observed? Also, explain why a left apex probe cannot be used for this SNV. The SNV sequence, on the strand shown in NCBI, and a few nucleotides adjacent to the SNV are below: 5'-------TGT(G/C)CAG------3'
Among the following statements regarding the use of viral vectors, which is (are) true: (can be more then one answer) A.Viral vector transfection has the defect of rapidly killing cells, given the high toxic potential of viruses. B.Generally, the effectiveness of a viral infection transfection is much higher than an electroporation or cationic liposome transfection. C.Even before infecting a cell, a virus can begin to express the indicators, accelerating the process of expression of the indicator in the culture medium. D.Adeno-associated viruses are little used in biophotonics, as they require simultaneous adenovirus infection.
