EXERCISE 1.3: MPN OF A SOIL SAMPLE (Groups of 2)
Materials

Flask with 100 mL of sterile water
100 mL sterile water
18 X 5 mL TSB
Sterile tubes
Method
1. Transfer 1g of soil to the flask containing 100 mL of sterile water. (This represents a 10-²
dilution) Shake on shaking platform for 10 minutes.
2. Allow the soil to settle for 10 minutes.
3. Prepare 104, 10-6 and 10-8 dilutions in water of the soil suspension in a final volume of 10 mL.
4. Perform an MPN count as indicated below in six sets of three tubes containing 5 ml of TSB.
5. Incubate at 37°C for 24 hours after which they will be stored at 4°C until your next lab period.
